PKC Levels in Serum and Bax/Bcl2,Caspase3 in Skeletal Muscle of Rats with Diabetes

Objective:To appraise the changes of PKC levels in serum and bax/bcl2,caspase3 in skeletal muscle of rats with diabetes.Methods:Sixteen SD rats were randomly divided into two groups:normal control group,diabetic group.We observed the general structure of the muscular tissue by HE sections.The expressions of PKC were studied by ELISA.The bax/bcl2 and caspase3 were studied by immunohistochemistry.Results:Histological observation:With HE staining,the skeletal muscle sections in control group showed normal structure.Morphometric changes of skeletal muscle in experimental diabetic rats were characterized primarily by widespread myoatrophy,muscle fiber denaturalization and necrosis,transverse striations of the fibers disappeared and interstitial fatty tissue hyperplasia.The level of PKC in diabetic group and normal control group was 110.60±5.94 and 74.72±7.19ng/ml(P0.01)respectively,the level of Bax was 26311±3698.30 and 16764±6116.10μm2(P0.05)respectively,the level of Bcl2 was 18573±8529.90 and 35533±485.52μm2 P0.05)respectively,the level of Caspase3 was 32419±3201.50 and 12123±1919.60μm2(P0.01)respectively.There were significant differences between two groups.Histological observation confirmed the injury of skeletal muscle caused by hyperglycemia and ischemia.The expression level of Bax and caspase3 increased significantly in diabetic rats than normal control group,the level of Bcl2 was lower than that of control group.Conclusion:PKC and apoptosis may be one of the causes of diabetic myoatrophy.
