Baslica uretim yeri bobrektir, fakat karaciger ve doku makrofajlan gibi ekstrarenal dokularda da eritropoietin sentezlenebilir. Doku hipoksisi eritropoietin uretiminin baslica uyaranidir. Hormonun sentezini kontrol eden hucreler kanin O2 kapasitesi, 02 basinci ve affinitesindeki degisikliklere cevap verirler. Ayrica lokal kan akimi ve hucrelerin 02 tuketimi de bir miktar rol oynayabilir. Eritropoietin CFU-E (Colony Forming Units-Erythroid) ve BFU-E (Burst Forming Units-Erythroid), hucre membranindaki spesifik reseptorleri etkiler ve boylece bu hucrelerin cogalmasini ve CFU-E hucrelerinin proeritroblasta donulmesini saglar. Abstract Erythropoietin: Structure, Metabolism and Mechanism of Effect Erythropoietin is an alpha-globulin whose molecular weight is 30400 Dalton. Its main production site is kidney, but little erythropoietin can also be synthesized in the extrarenal tissues sucfy as liver and tissue macrophages. Tissue hypoxia is essential stimulus of erythropoietin production. The cells controlling the synthesis of the hormone appear to respond to changes in the 02 capacity^ the 02 tension, and the affinity of the blood. In addition, the rates of the local blood flow and of the cellular 02 consumption may play some role. Erythropoietin affects the specific receptors in CFU-E (Colony Forming Units-Erythroid) and BFU-E (Burst Forming Units-ErythroId) celi membranes and therefore promotes proliferation of these cells and transforming of CFU-E to proerythroblast.
