Isolation,Identification and Enzymatic Characteristics of a Halophilic-amylase-producing Strain

A halophilic bacteria which produce extracellular amylase was isolated from the saline alkali soil of Shache,Xinjiang.Based on the characteristics of morphology,physiology,biochemistry and analyzing of 16S rDNA sequence,the strain was identified as genus of Halobacillus,and named Halobacillus sp.SCULCB HVA-10(GeneBank accession number: HQ620680).Studies on the condition of strains growth and enzyme producing had shown that: in high salt tolerance,growth was observed in 30%(ie 5.13mol/L) of salt concentration,and the optimum salt(NaCl) concentration for growth was determined as 10%-15%,so the strains were halophilic bacteria;in high alkali resistance,growth occurs at pH 6.0-11.0 with optimum growth at pH 7.0.There was a significant positive correlation between amylase activity and growth,Amglase actinty reached the highest of 600U when salt concentration ranging from 0% to 15%.
