Sitopaladi, a poly-herbal formulation inhibits IgE mediated allergic reactions

AIM: The present study is aimed to investigate the effect of Sitopaladi (SC-1), a polyherbal formulation, on IgE mediated allergic reactions both in vivo and in vitro. MATERIAL AND METHODS: SC-1 was evaluated by in vitro and in vivo methods for its effect on IgE mediated allergic reaction. RBL-2H3 cells were used for the in vitro study where β-hexosaminidase release was measured. Active paw anaphylaxis model of mice was used to study the activity in vivo. Cytokine levels were measured both in vitro and in vivo. The results obtained from in vivo study substantiated the in vitro data. LD50 for SC-1 was found to be more than 2000 mg/kg. RESULTS: Results from both in vitro and in vivo studies employing RBL-2H3 cell line and rodents respectively, revealed that SC-1 inhibits IgE mediated reactions significantly. CONCLUSION: These findings provide a rationale for the clinical use of SC-1 in IgE mediated allergic reactions. However, further mechanistic studies need to be performed.
