亚砷酸对HepG2肝癌细胞周期及P27、CycIinD1表达的影响

Objective To approach the mechanism of Arsenictrioxide affecting hepatic carcinoma cell cycle by vitro cell experiment by observing Arsenictrioxide influencing the level of CyclinD1 and P27 protein in the hepatic carcinoma cell. Methods Researching on Arsenictrioxide influencing the level of CyclinD1 and P27 protein in the hepatic carcinoma cell by immunohistochemistry SABC method and surveying the cells' cycle distribution by flow cytometer (FCM).Results The percents of P27 expressing cells are respectively (22.80±6. 94),(25.60±7.41),(28.00±5.21),(31.20±4.98) in the control group, low dose Arsenictrioxide group, median dose Arsenictrioxide group and high dose Arsenictrioxide group(By contrast to the control group , the difference all have statistical significance, P〈0.05 in the low dose group, the other two group are P〈0.01).The percents of CycinD1 expressing cells are respectively (38.40±5.68),(24.70±8.98),(18.90±6.51),(15.20±8.44)in the control group, low dose Arsenictrioxide group, median dose Arsenictrioxide group and high dose Arsenictrioxide group(By contrast to the control group, the difference all have markedly statistical significance, P〈0.01).The percents of G1 cells are respectively 66.5%, 70.6%, 73.4%, 77.5% in the control group, low dose Arsenictrioxide group, median dose Arsenictrioxide group and high dose Arsenictrioxide group. Conclusion Arsenictrioxide affect the HepG2 cell cycle maybe is through making the CyclinD1 declined and P27 protein rise. It target at the restriction point of G1-S, making the HepG2 cell cannot go through the restriction point of G1-S to proliferate and stoD the heDatic ceil on the G1 stage.
