Preparation of High Specific Polyclonal Antibody against Bifidobacterium longum

Objective To prepare the high specific polyclonal antibody against Bifidobacterium longum and provide a reference for immunological determination of Bifidobacterium longum.Methods The cell debris of Bifidobacterium longum obtained by ultrasonication was used as an antigen for immunization of mice to prepare antisera which was purified by the magnetic beads coupled with the same antigen,and determined for titer,purity and cross reactivity by ELISA,SDS-PAGE and dot hybridization respectively. Results The titer,recovery rate and purity of purified polyclonal antibody were about 1∶1 500,about 80%and 83%respectively.No nonspecific cross reaction of the purified polyclonal antibody with Staphylococcus aureus was observed.Conclusion High specific polyclonal antibody against Bifidobacterium longum was prepared,which might be used for detection of Bifidobacterium longum in commercial products.
