hydrochloride), an analog ofamantadine withsimilar activity and pharmacology, iscurrently being evaluated fortheprophylaxis andtherapy ofinfluenza A virus infection. Preliminary multiple-dose pharmacokinetics ofrimantadine havebeen examined ina fewstudies (1,2;M.C.NahataandM.T. Brady, Clin. Pharmacol. Ther.39:214, 1986). Thesemultiple-dose studies were notdefinitive, so no information regarding linearity, accumulation, orhalf-life was reported. Therefore, this study was conducted todetermine thesingleandmultiple-dose pharmacokinetics ofrimantadine inadults after 100mg administered onceandtwice daily (b.i.d.). Twenty-four healthy malevolunteers between theagesof 18and43yearscompleted thestudy. Thevolunteers demonstrated goodgeneral health asdetermined bythebase-line history, physical examination (including an electrocardiogram), andbase-line laboratory determinations consisting of a complete bloodcountwithdifferential, urinalysis, and serum chemistries. Thebodyweight ofeachsubject was within 10%ofideal bodyweight. Allsubjects gavewritten, informed consent toparticipate inthestudy.
