Combined effect of epidermal growth factor and cis-platinum for lung cancer xenografted tumor

Objective
To study the vombined effect of epidermal growth factor (EGF) and cis-platinum (CDDP) for lung cancer xenografted tumor, and preliminary mechanism.
Methods
Lung cancer xenografted tumor model was established. Nude mice divided into four groups (control group, EGF group, chemotherapy group, and EGF+ chemotherapy group). The lung cancer A549 cells were also tested for the ability to form tumors, tumor control rate, the expression of proliferation cell nuclear antigen (Ki-67), and the CD133 positive proportion in cancer cells.
Results
Tumor control rate of lung cancer xenografted tumor in EGF+ chemotherapy group was apparently higher than that in chemotherapy group, respectively 74.7% and 48.5%. The expression of Ki-67 in EGF+ chemotherapy group and chemotherapy group was respectively (45.38±7.98)% and (34.45±5.43)%, and there were more proliferating phase cancer cells in the former. The CD133 positive proportion of lung cancer xenografted tumor in EGF+ chemotherapy group was apparently higher than that in chemotherapy group, respectively (8.52±0.87)% and (4.19±0.56)%. As compared with control group and chemotherapy group, EGF+ chemotherapy group had evident difference (P=0.000, 0.031, 0.019).
Conclusion
EGF can enhance the sensitivity of cis-platinum to lung cancer cells, probably by inducing these cancer cells in stationary phase into cell proliferative phase.
