ISOLASI DAN UJI AKTIVITAS ANTIBAKTERI SENYAWA METABOLIT SEKUNDER DARI FRAKSI n-HEKSANA RANTING RUKAM (Flacourtia rukam)

Rukam plant (Flacourtia rukam) is one of the plants of the genus Flacourtia. Almost all parts of the rukam plant have been used traditionally for the treatment of various diseases, but no information has been found in the use of the branches. The purpose of this study was to isolate and determine the antibacterial activity of secondary metabolites from the n-hexane fraction of rukam twigs. The study began with the extraction of compounds by maceration using n-hexane as a solvent. The separation and purification of compound was carried out by chromatographic techniques. The structure of the compound was determined based on the analysis of spectroscopic data of FT-IR, 1H-NMR, 13C-NMR, and comparative literature. Antibacterial activity was determined by disc diffusion method using Escherichia coli and Staphylococcus aureus as test bacteria and determination of minimum inhibitory concentration (MIC) was carried out by micro dilution method. The isolated pure compound was in the form of white crystals (31 mg). Based on the analysis of spectroscopic data and comparative literature, the isolated compound belongs to the triterpenoid group, namely friedelin. This compound has been previously reported. Pure compound exhibits antibacterial activity against E. coli and S. aureus bacteria with a minimum inhibitory concentration value of 120 g/mL.
