COMPARISON BETWEEN FCA AND QUIL A IN IMMUNOPOTENTIATION OF RECOMBINANT FUSION PROTEIN GST-Sj32 VACCINATION IN MICE

Objective To compare the immunoenhancing effects between two adjuvants,Freunds complete adjuvant(FCA) and Quil A(purified saponin),in mice vaccinated by recombinant fusion protein SjGST-32(rGST-Sj32). Methods Mice were immunized three times with rGST-Sj32 plus FCA or Quil A . The worms and the liver eggs from the mice challenged with 40 cercariae of Schistosoma japonicum after 45 days were counted. Results As compared with PBS control group,the rGST-Sj32+FCA group and rGST-Sj32+Quil A group showed that the worm reduction rates was 42.9% and 46.0% respectively,the liver egg per gram(LEPG) reduction rate was 51.0% and 39.4% respectively, and the liver egg per female(LEPF)reduction rate as 23.9% and 8.5% respectively. And the specific antibody titers reached 1∶25 600 and 1∶51 200 respectively. Conclusion FCA or Quil A plus rGST-Sj32 could induce significant protective immunity in mice. However, the former induced significant anti-fecundity immunity, and the latter was not showing such an effect.
