The Joint Action of Nonylphenol and Soy Isoflavone on the Reproductive System of Male Rats

2007
To explore the combined adverse effects of nonylphenol and soy isoflavone on the male offspring reproductive in rat.[Methods] 50 pregnant rats were randomly divided into five groups (A:control group,B:nonylphenol group (200 mg/kg),C:nonylphenol+low soy isoflavone dose (50 mg/kg)group,D:nonylphenol+middle soy i soflavone dose (150 mg/kg)group,E:nonylphenol+high soy isoflavone dose (450 mg/kg) group).Different dose of soy isoflavone and nonylphenol were administered to pregnant rats in each group by garage(as a solvent, only peanut oil was delivered to control group)during the period from 13th to 19th day of pregnant.FI offspring(male)were selected and sacrificed on 90~(th) day and their reproductive system was examined.[Results]Compared with the control group, the activities of sperm, the sperm count in the tail of epididymis, the gonepoiesis quantity per day of the left testis, the weight and coefficient of testis,as well as ALP,LDH and TT in the group of soy isoflavone+nonylphenol administered rats decreased significantly.Compared with nonylphenol treated group, the weight and coefficient of testis, the gonepoiesis quantity per day,ALP and LDH indices of F1 male rats in the nonylphenol+50 mg/kg soy isoflavone treated group increased significantly;the weight and coefficient of testis, the gonepoiesis quantity per day,ALP and LDH increased significantly, but the activity of sperm, the quantity of living offspring and TT showed the trend of decrease in the nonylphenol+450 mg/kg soy isoflavone treated group.[Conclusion] Nonylphenol might cause hazards on the F1 male in differentiation period of reproductive system. The nonylphenol and 50 mg/kg soy isoflavone could show antagonistic effects, but the nonylphenol and 450 mg/kg soy isoflavone show additive actions on F1 male reproductive system of rats.
