Investigation of epigenetic modifier on HDAC1 and microRNA-410 expression in ovarian cancer cell lines

Abstract The purpose of this study was in vitro evaluation of 5-Aza-2′-Deoxycytidine (5-Aza-dC), Trichostatin A (TSA) and Cisplatin effects on HDAC1 gene and miRNA-410 expression in ovarian cancer cell lines and comparing with cotreatment results on gene expression. Influence of 5-Aza-2′-Deoxycytidine, TSA and Cisplatin on human cells was evaluated alone and together. We measured HDAC gene and miR-410 expression by Quantitative Real Time polymerase chain reactions (qRT-PCR) and the results were analyzed by SPSS and GraphPad Prism 7 software. While each drug effects the expression of HDAC1 and miRNA-410, their coadministration had more effective results. Cisplatin, TSA, and 5-Aza-dC have a great potential in terms of therapeutic agents in ovarian cancer patients; however, more studies are needed to optimize their specificity and effectiveness.
