The Protective Effect of Monomer Component from R. Giganteum(Schwaegr.) Par and Their Effect on Umbilicus Endothelial Cells and Its Inereasement of NO or NOS

To study the anti-atherosclerotic effect of 5 kinds of monomer composition from R.Giganteum(Schwaegr.) Par's,in vitro vascular endothelial cells were cultivated and the H2O2 were used to induce the oxidative stress injury model.The 5 kinds of chemical compound from R.Giganteum(Schwaegr.) were added to cultivated human vascular endothelial cells separately,and OD value was carefully determined by MTT method.The NO and NOS kit were used to detect the NO concentration and activity of NO in different groups.The most suitable stimulating concentration of H2O2 on HUVEC is 1mmol/L.The HVVEC injuride by H2O2 was co-cultured with different concentrations of the 5 kinds of chemical compound for 24 h.The inhibition rate indicates the difference significantly compared with the model group(P0.05 or P0.01).Therefore,the content of NO and NOS has significant difference(P0.05 or P0.01).Hence,the H2O2 concentration of 1 mmol/L could be used to establish the injured model of endothelial cell successfully.The 5 kinds of chemical compound have protective effect on ECV304 injured by H2O2,and could increase the activity of cNOS/iNOS as well as promote the synthesis of NO.
