Clinical study on the mecobalamin combined with pancreatic kallidinogenase for treatment of diabetic peripheral neuropathy

Objective:To observe the efficacy and safety of the treatment of diabetic peripheral neuropathy(DPN) with mecobalamin combined with pancreatic kallidinogenase.Methods:One hundred patients with type 2 diabetes were digital randomly divided into treatment group(n=50) and control group(n=50).The patients in treatment group were given mecobalamin tablet 0.5 mg tid and pancreatic kallidinogenase tablet 120 U tid.The patients in control group were only given mecobalamin tablet 0.5 mg tid.The course of treatment was six months.Hemorrheology,microalbumin in urine,β2-microglobulin in blood and urine,blood glucose,hemoglobin Alc,hepatic and renal functions,electromyogram and Doppler colour ultrasound of lower extremity artery were examined.Results:After 6 months of treatment,the effectiveness indexes which were better in mecobalamin combined with pancreatic kallidinogenase group than in mecobalamin only group were as follows:(1) Hematocrit values were(0.45±0.03) vs(0.45±0.04) before treatment,and(0.38±0.03) vs(0.42±0.04) after treatment in 2 groups(P0.05),respectively.(2) Microalbumin in urine was(65±14) vs(61±16) mg/mmol Cr before treatment,and(5.44±2.15) vs(15.47±3.10) mg/mmol Cr after treatment(P0.05),respectively.(3) Average wave amplitude of action potential was(129.6±39.4) vs(129.1±39.9) mV before treatment,and(79.7±18.5) vs(89.9±21.7) mV after treatment(P0.05),respectively.The differences of other indexes were not statistically significant between 2 groups.Compared with before treatment,the indexes in each group were significantly improved after the treatment(P0.05,P0.01).Conclusion:Mecobalamin only or combined with pancreatic kallidinogenase was effective and safe for treatment of DPN,and some indexes were improved more in treatment group than in control group.
