Expression of nuclear factor - kB and cyclo - oxgenase -2 in colonic adenoma polyps and colon cancer tissues and its relationship with angiogenesis

Objective To observe the expression of nuclear factor-κB (NF-κB) and cyclo-oxgenase 2 (Cox-2) in colonic adenoma polyps and colon cancer tissues, and explore its relationship with angiogenesis. Methods The expression of NF-κB p65, Cox-2 and CD34 in 35 cases of colonic adenoma polyp, 42 cases of colon cancer and 30 cases of normal colonic mucosa tissues was detected by immunohistochemistry. The correlation between the expression of NF-κBp65 and Cox-2 in colon cancer tissue and the correlation of them and microvascular density (MVD), clinicopathological features were analyzed. Results The positive expression rate of NF-κBp65 in normal colonic mucosa, colonic adenoma polyp and colon cancer tissues was respectively 30.0%, 57.1% and 81.0%, showing a trend of increase with the difference being statistically significant (P 0.05), and had statistically significant difference in the lymph node metastasis, liver metastasis and Dukes staging (P<0.05). The expression of NF-κBp65 had a significantly positive correlation with Cox-2 in colon cancer tissues (r=0.632, P<0.05). The number of MVD in colorectal tissues with NF-κBp65 and Cox-2 positive expression (34.1±10.7, and 28.5±9.8) was significantly greater than that in those with NF-κBp65 and Cox-2 negative expression (9.8±3.2 and 11.2±4.6) (P<0.05). Conclusion The NF-κBp65 and Cox-2 play an important role in the process of abnormal proliferation of colon cancer, invading the surrounding normal tissues and distant organ metastasis, and is closely related to the angiogenesis.
