Comparative study on extraction methods of genomic DNA from Momordica charnatia.

Objective To study the quality of genomic DNA from Momordica charantia extracted by different methods.Methods The total DNA was isolated from Momordica charantia by phenol-chloroform method,improved CTAB method and kit method,respectively.The quality of DNA was detected by Southern hybridization and PCR.Results The A260/A280 of the DNA extracted by improved CTAB method was 1.85 and the A260/A230 was 1.96.Otherwise,the hybridization signal could be detected in Southern blotting and the purpose strap could be amplified by PCR for the DNA extracted by improved CTAB method.Conclusion The improved CTAB method is the best method for the extraction of genomic DNA from Momordica charantia.
