Measurement of molecular conformation of immunoglobulin G developing for anti-neuron NMDAr1 antibody by atomic force microscopy

Objective To investigate whether IgG could be used as a label when binded with antigen of membrane protein ,and study the characteristics of conformation of anti-NMDAr1 polycolonal IgG. Methods AFM was used to detect images of anti-NMDAr1 polycolonal IgG affiliated on mica under physiological environment in order to determine the characteristic 3D structure of it.Results The images and data showed that the anti-NMDAr1 polycolonal IgG has characteristic structure described as an elliptical sphere of 136.4?× 62.8?×26.1?.On section, there were two peaks with the width of 13 nm at the edge of ellipse. Conclusion Atomic force microscopy is a new method which is capable to provide images at a nanometer scale resolution. It could be used to study the structure of IgG.IgG could be used as a label in the study of membrane receptor protein.
