Proliferation characteristics of culturing coxsackievirus strain YY157 in various cell lines

2015
Objective To investigate the growth and proliferation features of coxsackievirus A16 (Cox A16) strain YY157 cultured in different cell lines, and provide a reference for the research of vaccine. Methods Cox A16 strain YY157 was inoculated into Vero, MRC-5 or KMB17 cells and adapted by passage culturing. Meanwhile, cytopathic effect (CPE) was observed, with infective titer of the virus determined and its multiplication curves drawn. Results Cox A16 strain YY157 could induce CPE in all three cell lines. In Vero cells, CPE appeared early and the virus propagated rapidly with its titer reached 8.50 lgCCID50/mL. In MRC-5 cells, the virus proliferated slower than that in Vero cells with its titer reached 7.5 lgCCID50/mL. In KMB17 cells, the growth rate of the virus was relatively lower with its titer only 7.0 LgCCID50/mL. Conclusions Cox A16 strain YY157 is able to infect and propagate in Vero, MRC-5 and KMB17 cell lines. However, the sensitivity of those cell lines to the strain and the proliferation rates of the virus in those cell lines are different.
