Effects of curcumin on interleukin-6 and tumor necrosis factor-α in APP/PS1 transgenic mice.

Objective To investigate the effects of different concentrations of curcumin on tumor necrosis factor-α(TNF-α)and interlukin6 (IL-6)in APP/PS1 transgenic mice.Methods A total of 45 female 6-month-old mice were divided into 3 groups:control group(wildtype mice,n =9),model group(APP/PS1 transgenic mice,n =9),and curcumin group(APP/PS1 transgenic mice treated with curcumin,n=27).The rats of control group and model group were housed under ad libitum feeding conditions.The rats of Curcumin group were administrated with curcumin orally (60,160,600 mg·kg-1).Three mice of each group were sacrifced at 9,12 and 18 months of age,and the brain and serum samples were harvested to analyse the levels of TNF-α and IL-6 by ELISA.Results Compared with the wild-type mice,the levels of TNF-α and IL-6 in the brain and serum of APP/PS1 transgenic mice began to increase from 9 months of age,and the significant differences were found at 12 and 18 months of age(P 0.05,P 0.01).After the treatment of curcumin(60,160,600 mg·kg-1),the levels of TNF-α and IL-6 in APP/PS1 transgenic mice degraded dose-dependently.Conclusion With the development of AD,the levels of TNF-α and IL-6 in the brain and serum of APP/PS1 transgenic mice increased gradually.After the treatment of curcumin,the levels of TNFα and IL-6 in APP/PS1 transgenic mice degraded dose-dependently.
