Survivin及其剪接变异体survivin-△Ex3,survivin-2B mRNA在乳腺癌中的表达和临床意义

Objective To investigate the expression of survivin and its splicing variants survivin-△Ex3 and survivin-2B in human breast cancer tissues and its clinical significance. Method With reference to the expression of GAPDH, the expression of survivin and its splicing variants survivin-△Ex3 and survivin-2B mRNA was determined by both reverse transcription-polymerase chain reaction (RT-PCR) and semi-quantitative electrophoresis in 50 breast cancer and adjacent breast tissues. The protein expression of estrogen receptor (ER), progesterone receptor (PR), and Her-2 gene was determined by immuno-histochemical method. Result The expression of survivin and its splicing variants survivin-△Ex3 and survivin-2B mRNA in the breast cancer tissues was significantly higher than the adjacent normal breast tissues (P<0.05). The high level of expression of survivin-△Ex3 and survivin-2B mRNA was correlated with the axillary lymph node metastasis (P<0.05). Conclusion Survivin and its splicing variants survivin-△Ex3 and survivin-2B may be used together with other prognostic markers in the prognosis and therapy of breast cancer.
