Effect of homocysteine on mitochondrial membrane potential and reactive oxygen species of human umbilical vein endothelial cell

2011
Objective To investigate the effects of reactive homocysteine (Hcy) on the mitochondrial membrane potential (JC-1) and reactive oxygen species (ROS) production in human umbilical vein endothelial cell (HUVEC). Methods HUVEC were isolated and cultured , treated with Hcy in different concentration(0mmol/L,0.01 mmol/L,0. 1 mmol/L,1 mmol/L,3 mmol/L,5 mmol/L) and for different duration(3 h,6 h,12 h,24h) , JC-1 and ROS were detected with the flow cytometry. Results The JC-1 decrease(F = 101. 82,F = 85.257,P<0.01) and ROS increase (F = 89. 82,F = 82. 235,P <0. 01), had a significant difference. Conclusion Hcy could induce HUVEC injury by decreasing MMP and producing ROS in a time- and dose-dependent manner.
