Norcantharidin disturbed cell cycle progress in cancer HL-60 cells and its mechanism.

Objective To confirm the influence of norcantharidin(NCTD) on cancer cell cycle,and to further explore the anti-cancer mechanism of NCTD.Methods HL-60 Cells were cultured and treated with NCTD,3H-TdR incorporation assay was performed to reflect DNA replication,flow cytometry was used to detect cell cycle progress,and the cell-cycle-regulation proteins were detected by Western blotting.Results After treatment with NCTD for 12 h,DNA replication was significantly inhibited and the cell cycle was arrested mainly at S phase in a dose-dependent manner.G2/M Phase accumulation was also observed and G2/M regulation proteins Cdk1,Cyclin B1,and Cdc25c were down-regulated.ConclusionNCTD could interfere with cancer cell cycle through inhibiting DNA replication and deregulating cell-cycle-regulation protein.
