Effect of the Gui Long Jian Gu Prescription on expression of Bcl-2 and Fas protein in apoptosis of rat osteoblasts induced by TNF-α

Objective: To investigate effect of the Gui Long Jian Gu Prescription on expression of Bcl-2 and Fas protein in apoptosis of rat osteoblasts induced by Tumor Necrosis Factor-α.Methods: Primary cultured calvaria osteoblasts of rats were divided into normal control group and traditional Chinese herbs group.The osteoblasts of traditional Chinese herbs group were fed the drug serum of the Gui Long Jian Gu Prescription,and the osteoblasts of normal control group were fed fetal bovine serum.After 7 days,these cells were incited with TNF-αof 30ng/mL.Flow cytometry was used to evaluate apoptosis,and the expression of Bcl-2 and Fas protein were measured by immunohistochemical ABC method with imaging analysis process.Results: The expression of Bcl-2 was increased and the expression of Fas was not obvious in traditional Chinese herbs group.The expression of Fas was increased and the expression of Bcl-2 was not obvious in normal control group.In the apoptosis of osteoblasts,the rates of traditional Chinese herbs group at early stage and advanced stage were lower than the rates of normal control group,there is a statistically significant difference between two group(P0.05).Conclusion: The expression of Bcl-2 is up-regulated by the Gui Long Jian Gu Prescription,but the expression of Fas was not obvious.To some extent,the Gui Long Jian Gu Prescription can inhibit apoptosis of osteoblasts.However,whether the proliferation of osteoblasts was worth researching in future.
