Polymorphism of apolipoprotein E, methylenhydrofolate reductase and paroxonase genes in patients with Alzheimer`s disease

Ethiology of Alzheimer's disease has not been completrely discovered until today. Apolipoprotein E gene and his epsilon 4 allele is one of identified risk factors for DAT. This study included 187 healthy subjects and 80 DAT patients. PCR-RFLP method was used to investigate frequency of alleles and genotype distribution for Apo E. Apo E e 4 allele was present in 35, 7% patients and only in 3.8% of healthy subjects. Difference was statistically significant in MTHFR genotype distribution between heakthy subjects and patients, however, allele frequencies did not differ significantly. Subgroup of patients were analysed. Conclusions: This preliminary results showed significant difference in Apo E and MTHFR genotype distribution, as well as Apo E e4 allele, but not in PON1 genotype, between patients and healthy subjects. In patients with Apo E e4/e4 genotype, only PON1 genotype was shown to be additional risk factor dor DAT, compared to healthy subjects.
