Effect of JAB1 down-regulation on LPS-induced TNF-α and IL-6 in mouse macrophage cells

Objective To study the effect of down-regulation of JAB1 in the inflammatory reaction.Methods The recombinant plasmid pPUR/U6-siJAB1 was constructed and transfected into RAW264.7 cell to down-regulate JAB1.The concentrations of TNF-α and IL-6 in the cell culture media were detected by ELISA.Results The recombinant plasmid pPUR/U6-siJAB1 was obtained successfully.The expression of JAB1 in RAW264.7 cell was markedly decreased.The concentrations of TNF-α and IL-6 stimulated by LPS in the cell culture media were lower than the normal group significantly.Conclusion Low expression of JAB1 in RAW264.7 cell degrades the production of inflammatory cytokines,and JAB1 may exert powerful effect on pro-inflammatory reaction caused by LPS.
