Gene function encoding TLR7 recognizing the synthetic compound having an immunopotentiating effect of such imidazoquinoline compound is deficient on its chromosome, there is provided a synthetic compound non-human animal model unresponsive. All or part of the gene fragment of the gene site containing an intracellular domain and a transmembrane region of TLR7 gene obtained from a mouse gene library, to build a targeting vector by replacing the plasmid with the poly A signal and a marker gene , and introduced into embryonic stem cells after the targeting vector was linearized, the target embryonic stem cells lacking TLR7 gene function, microinjected to produce a chimeric mouse blastocysts of mice, wild-type mice and the chimeric mice mated the door to prepare a heterozygous mice to produce a TLR7 knockout mice by intercrossing such heterozygous mice.
