Changes of Smurf2 and SnoN expression in kidney tissues of type 2 diabetic rats and their biological roles

Objective To observe the changes of Smad ubiquitin regulatory factor 2(Smurf2) and transcription co-repressor SnoN expression in renal tissues of type 2 diabetic rats and their biological roles.Methods SD rats were injected with STZ after taking high fat and high sucrose diet for 12 weeks,and were killed thereafter at the 8th,12th,and 16th weeks respectively.SnoN,Smurf2,TGF-β1,and Smad2 in renal cortex were detected by immunohistochemistry and Western blotting.SnoN mRNA was checked with RT-PCR.Results The expression of Smurf2 in diabetic renal tissues was significantly increased(P0.01),meanwhile the expression of SnoN was significantly decreased(P0.05),and there was a negative correlation between these two proteins(r=-0.71,P0.01).SnoN mRNA in diabetic renal tissues was unchanged among groups(P0.05).Conclusion The results suggest that the downregulation of SnoN expression may be mediated by Smurf2 in type 2 diabetic nephropathy.
