Association of complex karyotype with inferior progression-free and overall survival in mantle cell lymphoma.

7565Background: Complex karyotype (CK) has been associated with inferior outcomes in mantle cell lymphoma (MCL) and other hematologic malignancies, including, but has only been evaluated in small MCL cohorts. We conducted a multi-center assessment of the impact of a CK on progression-free (PFS) and overall survival (OS) in patients with previously untreated MCL. Methods: Untreated patients (pts) with MCL from 4 academic centers diagnosed between 1993 and 2015 were included. Pts with 3 or more chromosomal abnormalities were defined as having CK. We described differences in clinical variables of interest between pts with and without CK and used the Kaplan-Meier method to estimate OS and PFS for each group. Multiple variable (MV) Cox regression analyses including MIPI components and demographics were performed to identify independent predictors of PFS and OS.
