Development of a Novel Nanoparticle Formulation of Cisplatin to Improve Chemoradiation Therapy

HBI also induced a significant systemic inflammatory reaction as demonstrated by increased plasma levels of IL-6 at four hours and KC/CXCL1 at four hours, as well as 1-3 days (p<0.05). Radiation also increased tissue levels of IL-1 and KC/CXCL1 in the small intestine and IL-6 and KC/CXCL1 in lung tissue (p<0.05). Depletion of Treg cells with PC61 prior to HBI did not significantly change the levels or patterns of cytokine and chemokine responses to irradiation. Treg cells depletion also did not influence the repair of radiation-induced intestine damage as assessed by histology. Conclusions: We have developed a model of normal tissue injury to analyze the interactions of radiation with Treg cell depletion. The data suggest that the depletion of Treg cells does not influence HBIinduced inflammatory response or tissue injury. This model can be used to test the potential deleterious consequences of immunemodulating drugs in combination with irradiation prior to their clinical evaluation. Author Disclosure: S. Ma: None. J.
