Construction of Newcastle disease virus-vectored canine distemper virus(CDV) vaccine and induction of CDV neutralizing antibody

In order to develop a safe and efficient canine distemper vaccine based on Newcastle disease virus(NDV) La Sota vaccine strain,we constructed and rescued a recombinant NDV which expressed hemagglutinin(H) protein of canine distemper virus(CDV,Rockborn-20/8 strain),named rLa-CDV-H,by reverse genetics techniques.The expression of CDV H protein in recombinant virus was confirmed by western blot and indirect immunofluorescence assay.rLa-CDV-H retained its high-titer growth property and low pathogenicity in embryonated chicken eggs.Moreover,rLa-CDV-H induced significant CDV neutralizing antibodies in 12 week old dogs post-inoculation.This result indicated that rLa-CDV-H was able to induce efficiently immune responses in dogs,which showed that rLa-CDV-H had potential of being a novel CDV vaccine.
