Nuclear transfer in rabbits

Blastomeres of rabbits from cell-16 to cell-32 stage were used as the donors for muclear transfer in this study. The blastomere was injected to the preivitelline of enucleated oocyte by micromanipulation. As the result, the fusion rate was 76.8% after twice electric stimulus for the 229 recombined embryos. The cleavage rate (65.7%, 23/25) and the rate developed to morula and blastocysts (22.9%, 8/35) for recombined embryos were significantly increased (P<0.01) by co-culture of fetal fibroblasts compared with droplet direct culture (43.3%, 13/30;16.7%,5/30) respectively. Nine estrus synchronized recipients were transplanted 111 reconstructed embryos into their oviducts, two recipients were in pregnancy and one successfully delivered two cloned offspring after full period of pregnancy.
