Method for detecting a target nucleic acid, said process comprising contacting the template nucleic acid from a sample with (i) a signaling system (ii) a primer nucleic acid tail having a connecting zone to the template comprising and the tail comprising a linker segment and a zone of target binding in the presence of appropriate nucleoside triphosphates and an agent for polymerization, under conditions such that the area of template binding primer will hybridise to a complementary sequence in the template nucleic acid and be extended to form a product of primer extension, separating any such product from the matrix, after which the connecting region to the target in the primer tail will hybridize to a sequence in the amplification product primer corresponding target nucleic acid with and wherein any specific hybridization of the target produces a detectable change, and n signaling system, so that the presence or absence of target nucleic acid in the sample is detected relative to the presence or absence of a detectable change in the signaling system.
