Bioinformatic characterization of recombinant pseudorabies virus PGO expressing ORF2 gene of porcine circovirus type 2

Transfer plasmid PGO and genome of PRV attenuated vaccine were transfered into ST cells with transfection techology and the recombinant PRV was obtained with three times of plaque assay.Research on partial biological characteristics of the recombinant virus showed that the TCID50 of the recombination virus on different cell lines is PK 104.125/0.1,ST 106.125/0.1,VERO 104.625/0.1,IBRS-2 106.25/0.1mL;physicochemical characters on the recombinant virus indicated that it is the same as normal PRV.RT-PCR,inverted fluorescence microscope and IPMA were applied to identify the expressing of the inserted gene in the recommbinant virus.Safety testing indicated that the immunization of the recommbinant virus was safe for mice;genetic stability showed that the inserted ORF2 of PCV2 played an excellent character.This study laid foundation for the research and development of recombinant pseudorabies virus vaccine expressing ORF2 gene of PCV2.
