Expression and Prognostic Value of E-cadherin,p27 and Proliferating Cell Nuclear Antigen in Gastric Carcinomas and Matched Lymph-node Metastases

2003
Objective To study the expression and prognostic value of E-cadherin(E-cd),p27 and proliferating cell nuclear antigen (PCNA) in gastric adenocarcinoma and their lymph-node metastases. Methods Immunohistochemical method was used to detect E-cd,p27 protein and PCNA expression in 20 normal gastric mucosa tissues,20 gastric epithelial dysplasia,109 advanced gastric adenocarcinomas and matched lymph-node metastases. Results The positive expression rates of E-cd,p27 protein and PCNA in gastric epithelial dysplasia, gastric adenocarcinomas and matched lymph-node metastases were 85%(17/20),40.37%(44/109),23.85%(26/109),60%(12/20),33.03%(36/109),19.27%(21/109) and 25%(5/20),74.31%(81/109),77.98%(85/109) respectively with significant difference,P0.01.The positive expressions of E-cd and p27 were closely related to Lauren type,TNM categories, and prognosis. The expression of PCNA was closely correlated with growth pattern,Lauren type, blood vessel invasion, and prognosis. The expressions of E-cd and p27 were closely linked to diffuse-type gastric adenocarcinomas.Comparing the primary cancers with the matched lymph-node metastases, the expressions of E-cd and p27 were reduced. Conclusions E-cd,p27 and PCNA might be valuable markers in assessing biological behavior and prognosis in gastric adenocarcinomas.
