Isolation of Hantavirus from Apodemus Peninsulae and Analysis of Its S Fragment

Objective To investigate phylogenetic position of hantavirus in Apodemus peninsulae from Jilin province. Methods Viral isolation, RT-PCR, Cloning and sequenceing were used to investigate hantavirus of Apodemus peninsulae. Results Two strains were isolated from 4 positive lung samples of Apodemus peninsulae, namely hantavirus CJI89 and CJI93. The S fragment of Hantavirus CJI93 was cloned, sequenced and analyzed.The genotype of CJI93 is HTN. The nucleotide sequences of the S fragment of CJI93 showed ~94.3% identity with that of strain Aa2499 (the highest homogoly), ~83.3% identity with that of strain Ap61 and ~82.9% identity with that of strain of Ap63 (the least homology). Phylogenetic analysis also indicated the closest relationship between CJI93 and Aa2499, which is hantavirus of Russia Apodemus agrarius, closer relationship between CJI93 and HTN hantavirus in China and the farthest relationship between CJI93 and Ap61, Ap63, which are hantavirus of Russian Apodemus peninsulae. Conclusion Results showed the strain CJI93 isolated from Jilin province of China is one of the new subtypes of HTN virus.
