Validated HPTLC Method for Determination of Sugars in Diploid (2x) and Tetraploid (4x) Cytotype of Physalis angulata L., An Important Medicinal Plant

AbstractA rapid, simple, improved and precise high performance thin layer chromatography method has been developed for the comparative estimation of sugars in two cytotypes i.e., 2x and 4x of crude plant part extracts of Physalis angulata, an important medicinal plant. Chromatographic separation was achieved by employing a mobile phase consisting 1-propanol: ethyl-acetate: water (3:1:0.1 v/v/v) on pre-coated Silica gel 60F254 plates and derivatized using DPA method (diphenylamine). Densitometry analysis was carried out at 600 nm in adsorption reflection mode. A significant difference in the quantity and quality of analyzed sugars was detected among both the cytotypes. The method was shown to be sensitive, accurate and reproducible for the determination of sugars in plants.
