AVALIAÇÃO DA CITOTOXICIDADE, MUTAGENICIDADE E ANTIMUTAGENICIDADE DO EXTRATO ETANÓLICO DE Gochnatia polymorpha

No Brasil muitas plantas sao utilizadas na Medicina popular para o tratamento de doencas, dentre elas encontra-se Gochnatia polymorpha . O presente estudo teve por objetivo descrever os efeitos do extrato etanolico da Gochnatia polymorpha (EEGP) sobre o DNA da cebola e em celulas de melanoma murino B16F10 por meio das tecnicas do ensaio do MTT e Allium cepa . Os resultados quantitativos indicaram que o EEGP e citotoxico para celulas de melanoma murino nas concentracoes entre 1000 e 31,25µg/mL. O EEGP nao possui atividade mutagenica no ensaio de Allium cepa quando testado na concentracao de 15x10 -6 µg/mL, e esse mesmo extrato ainda apresentou porcentagem de reducao de danos no DNA de 43,90% quando associado ao metilmetanosulfonato. O EEGP apresentou atividade antiproliferativa e citotoxica, o que indica atividade quimioterapica em B16F10. Tambem apresentou atividade antimutagenica o que indica capacidade de prevenir lesoes no DNA que podem se correlacionar ao desenvolvimento de tumores. ABSTRACT In Brazil, several plants are used in folk medicine to treat diseases, among them, is the Gochnatia polymorpha. This study aimed to evaluate the effect of an ethanolic extract of Gochnatia polymorpha (EEGP) on chromosomal aberrations induced by methyl methanesulfonate (MMS) in cultured meristematic cells of Allium cepa and also in murine melanoma cells (B16F10) by MTT assay and Allium cepa. Quantitative results indicated that the EEGP is cytotoxic to B16F10 cells at concentrations from 1000 to 31.25 ?g/mL. Data of Allium cepa assay showed that EEGP did not have mutagenic activity at the concentration of 15x10 -6 ?g/mL, however, it was verified a reduction of 43.90% in damage when EEGP was associated with MMS. EEGP have antiproliferative and cytotoxic activity, indicating chemotherapeutic activity in B16F10 cells.
