Study on the gene polymorphism of intercellular adhesion molecule-1 in type 2 diabetes mellitus patients with retinopathy

Objective To study the distribution frequency of intercellular adhesion molecule-1 gene 469K/E polymorphism and its relationship with type 2 diabetes mellitus patients with retinopathy. Methods The genotype frequencies of intercellular adhesion molecule-1 gene 469KK,KE and EE,and the levels of the fasting blood glucose,triacylglycerol,glycosylated hemoglobin,plasma free intercellular adhesion molecule-1 and total cholesterol were detected with PCR in 102 patients with diabetic retinopathy and compared the changes with 120 patients without diabetic retinopathy and 150 healthy controls. Results The levels of fasting blood glucose,glycosylated hemoglobin,total cholesterol,triacylglycerol,low density lipoprotein cholesterol and plasma free intercellular adhesion molecule-1 were significantly higher and the level of high density lipoprotein cholesterol was significantly lower in patients with type 2 diabetes mellitus than those in health controls(P0.01). The levels of total cholesterol,triacylglycerol and plasma free intercellular adhesion molecule-1 were significantly higher in patients with diabetic retinopathy than those in patients without diabetic retinopathy(P0.01). The genotype distribution frequencies of 469KK,KE and EE in three groups were statistically different (χ2=7.915,P0.01). The distribution frequency of allele E was significantly higher in patients with diabetic retinopathy than that in healthy controls and patients without diabetic retinopathy(P0.05). In patients with type 2 diabetes mellitus,the level of plasma free intercellular adhesion molecule-1 in allele 469E of intercellular adhesion molecule-1 carriers was significantly higher than that in allele K carriers (P0.01). Conclusion The diabetic retinopathy is related to the lipid metabolism disorder and the 469 KE gene polymorphism of intercellular adhesion molecule-1.
