The invention discloses application of ilicis routundae cortex acid derivatives in preparing an antitumor drug. Ilicis routundae cortex acid is obtained by extracting, separating and purifying ilicis routundae cortex and used as a lead compound, the ilicis routundae cortex acid is first reacted to generate ilicis routundae cortex acid esters, then the ilicis routundae cortex acid esters react with succinic anhydride to generate succinyl ilicis routundae cortex acid esters, and finally the succinyl ilicis routundae cortex acid esters react with amino acid methyl ester to prepare a series of derivatives. The pharmacological experiment proves that an amino acid derivative of the succinyl ilicis routundae cortex acid esters has antitumor activity and has an effect on inhibiting prostatic cancer cells, lung cancer cells, liver cancer cells, cervical cancer cells and gastric cancer cells, and the inhibiting effect of the derivative on the tumor cells is better than nuclear parent compound ilicis routundae cortex acid.
