Determination of Phosphatidylcholine and Lysophosphatidylcholine in Soybean Phospholipids by HPLC

2011
OBJECTIVE To establish an HPLC method for simultaneously determining phosphatidylcholine (PC), lysophosphatidylcholine (LPC) in soybean phospholipids. METHODS The HPLC system consisted of a Phenomenex silica column(4.6 mm×150 mm, 5 μm), a mobile phase of n-hexane-isopropanol-water(5∶80∶20). The flow rate was 0.7 mL.min-1. Column temperature was set at 25 ℃ and the wavelength of detector was set at 206 nm. RESULTS The calibration curve was linear in the range of 0.4-2.4 mg.mL-1 for PC and 10-60 μg.mL-1 for LPC, respectively. The average recovery for PC was 99.4% and RSD was 0.40%(n=6). The average recovery for LPC was 98.0% and RSD was 1.65%(n=6). CONCLUSION The method is simple, rapid and specially. It can be used for the determination of PC and LPC in soybean phospholipids.
