The invention discloses a method for preparing a multiple-structure multi-stimuli-responsive hydrogel layer-by-layer assembled film. The method comprises the following steps: (I) preparing a working electrode; (II) assembling a (PAH/PAA)m layer-by-layer assembled film on the surface of the working electrode PG; (III) assembling a (PAA-PBA/Dex)n layer-by-layer assembled film on the surface of the PG/(PAH/PAA)m electrode; (IV) assembling a PDEA (polydiethylene glycol adipate) hydrogel film on the surface of the PG/(PAH/PAA)m/(PAA-PBA/Dex)n electrode; and (V) assembling a PNIPAm (poly(n-isopropyl acrylamide)) hydrogel film on the surface of the PG/(PAH/PAA)m/(PAA-PBA/Dex)n/PDEA electrode. The method has the beneficial effects that: (1) the method is simple and convenient without complex instrument equipment; (2) two different methods for preparing the film are combined to successfully construct a hydrogel layer-by-layer assembled film having a unique dual structure; and (3) the film has the stimuli-responsive property to the pH value of the environment, fructose molecules, salt, temperature and methanol.
