Detection of ion channels of in vitro cultured neural stem cells before and after the differentiation in seahorse

objective: To study the change of voltage dependent Na+, K+and Ca2+channels before and after the differentiation of neural stem cells. Methods:Whole-cell recording technique was used to detect voltage dependent Na+current, Delayed rectifier K+current, voltage dependent Ca2+current at 3d,7d,14d and 21d before and after the NSCs differentiation. Results: INa failed to be detected before and after the NSCs differentiation. The percent of IKdetected was 11. 76%, 24. 75%, 46. 51% and 76. 18%, respectively at 3d, 7d, 14d and 21d after differentiation. The amplitude of ICa at any time recorded was (119 ± 73)pA, (213±98)pA,(324±151)pA,(735±312)pA and 1079±307)pA before and after the differentiation. Conclusion: INa failed to be detected before and after the differentiation. Both the percent of IKdetected and the amplitude of IN. increases when the time of differentiation became longer.
