Sterilization for Sorting

In order to aseptically sort and collect cells for tissue culture work, it is imperative that thorough cleaning and sterilization procedures be implemented before and after the use of Fluorescent Activated Cell Sorter instruments. The intent of sterilization is to use an agent that effectively destroys all microorganisms (e.g., viruses, bacteria, fungi and their spores) on inanimate surfaces. The following protocol addresses the steps and reagents necessary to achieve sterilization. Adequate and routine cleaning is a prerequisite for any sterilization procedure. The exposure time of the microorganism and spores to the sterilant agent will result in greater likelihood of elimination. Generally, 10 minutes is sufficient time to destroy most organisms. However, in the case of cell sorter instruments, thin tubing, narrow channels and other areas that are generally difficult to access may harbor organic materials in addition to microorganism. It may therefore be desirable to lengthen the exposure time to 20 or 30 minutes.
