Buffalo Follistatin cDNA cloning and sequence analysis

Follistatin(FS) affects animal reproductive mechanisms through inhibiting secretion of Follicle-stinmulating hormone(FSH) in paracrine and autocrine.The objective of this study is to elucidate the low reproductive ability in the Bubalus bubalis by analysis FS cDNA sequence.Primers were designed based on the Bos taurus FS cDNA released in GenBank.The purpose segment in RT-PCR technology was amplified and the target gene was sub-cloned to pMD-18T vector before sequencing.The results indicated that this study had successfully obtained FS cDNA from Bubalus bubalis.Homologous and evolutional analysis showed that FS had a high conservation.It was also found that there were two mutants of amino acids in this study,which may impact the normal biological function of FS,and in turn,affects the reproductive abilities in Bubalus bubalis.
