Analysis of cell-tissue grafts under weightless conditions using confocal fluorescence microscopy

ABSTRACT The results of studies of viable cells in the control cell, composed of tissue transplant using confocal laser microscopy, fluorescence at wavelengths of 488 nm and 561nm fluorophore using propidium iodide (propidium iodide, PI SigmaAldrich USA). Processing of the images was carried out using the software ANDOR. It is experimentally shown that the method of confocal fluorescence microscopy is one of the methods for detecting cell information populated in 3-D bio-carrier with a resolution of at least 400 nm. Analysis of the micrographs suggests thatthe cells were part bio-carrier for 30 days in experiment synchronous ground retained their viability compared to thespace in which the cells in the composition bio-carrier hardly detected. INTRODUCTION Since the beginning of space exploration conducted research aimed at the study of space flight factors on the processes of life, development, differentiation, and repair damaged structures of living organisms 1,2 . During this time, have been used a variety of biological models (studies in vivo), which allowed to find a number of important laws 3-6 . Currently, the useof cell, crops for space research is quite advanced from the position of bioethics and informative, since the objects occupy small volumes and do not require complex life support systems. Data were obtained that microgravity alters themorpho-functional state of the cells in vitro, their proliferative activity, gene expression and other intracellular processes. These effects are particularly pronounced in mechanosensitive differentiated cells such as osteocytes, fibroblasts, myocytes, endothelial cells 7-11 which manifest disorders of the musculoskeletal system in astronauts 12 . Study of the influence of various physical factors of the space environment (radiation, hyper and microgravity, etc.) on cell culture of hondro and osteoblastic differons is a promising and relevant. In IEMb of Samara Medicine University established himself biological model to study the effects of cosmic factors on cell cultures settled in the 3D- bio-carrier, which is represented by allogeneic demineralized Spongiosis 13 ,14 .
