Engineering bacteria based on chloride peroxidases and realization method thereof

2014
The invention relates to an engineering bacteria based on chloride peroxidases and a realization method thereof, and relates to the technical field of gene engineering. The realization method comprises the following steps: carrying out PCR (Polymerase Chain Reaction) amplification on streptomyces griseorubens genome DAN taken as a template with a primer which contains an enzyme cutting site to obtain a chloride peroxidase encoding gene; then inserting the chloride peroxidase encoding gene into an escherichia coli expression vector pET-22b(+) to obtain a recombinant expression vector which contains the chloride peroxidase encoding gene; then leading the recombinant expression vector into a escherichia coli expression strain, and screening to obtain the engineering bacteria of chloride peroxidases. The method disclosed by the invention can be used for realizing in-vitro large-quantity expression and synthesis of the chloride peroxidases and finally realizing the fast degradation of lignin by applying gene engineering means aiming at the defects that the application range and effect of the chloride peroxidases are seriously restricted because the chloride peroxidases are intracellular enzymes mostly in vivo and lower in expression quantity and the like in the prior art.
