2015
This study aims to determine the effect of ethanol extract of black cumin seeds (EEBJH) of the phagocytic activity and the secretion of reactive oxygen intermediates (ROI) murine peritoneal macrophages infected L.monocytogenes Switzerland. The design of the study was experimental design with a control group. Seventy two male mice of Swiss strain weighing between 20-30 grams were used. Mice were divided into 6 groups, each of 12 heads. Group 1 (negative control group), was given distilled water orally. Group II (positive control group), the tested animals were given orally imboost. Group III, IV, V, and VI, as the treatment groups, respectively - each given EEBJH with a dose of 1, 5, 25, and 125 mg / kg body weight / day orally for 14 days. On day 15, all mice infected L.monocytogenes. Peritoneal macrophage phagocytic activity was observed by the method of latex while ROI secretion activity was observed by the method of nitro blue tetrazolium (NBT) reduction assay. Do different test average phagocytic activity and the secretion of ROI between groups by ANOVA followed by LSD test with a 95% confidence level. The results showed that administration of EEBJH increase ROI phagocytic activity and the secretion of peritoneal macrophages infected by L. monocytogenes. The mortality rate of tested animals in the treatment group was lower compared to those of the negative group. Phagocytic activity of macrophages and secretion of ROI was highest on day 14. Phagocytic activity of macrophages and secretion ROI was highest in the group MBJH receiving dose 5mg/kg/day. Based on research results, it can be concluded that EEBJH has the effect of increasing the activity of macrophage phagocytosis and ROI secretion of Swiss mice infected with L. monocytogenes.
