Multiplication of Dendranthema zawadskii ssp. latilobum var. leiophyllum through Multiple Shoot Induction from Rhizome-Derived Shoot Tip Culture

Direct multiple shoots were induced in shoot tips derived from in vivo rhizomal buds of Dendranthema zawadskii ssp. latilobum var. leiophyllum on Murashige and Skoog (MS) medium supplemented with 6-benzylaminopurine (BAP) alone at 0.5-3.0 ㎎ㆍL?¹ or combinations of 0.5-3.0 ㎎ㆍL?¹ BAP and 0-0.5 ㎎ㆍL?¹ α-naphthalene acetic acid (NAA). A combination of 0.5 ㎎ㆍL?¹ BAP, 0.01 ㎎ㆍL?¹ NAA, and 30 gㆍL?¹ sucrose was optimal for multiple shoot formation (45 shoots per shoot tip) without roots within 4 weeks. Subculture of these shoots on the same medium resulted in similar reponses in multiple shoot formation within 4 weeks. Of the four basal media (1/2MS, MS, 2MS, B?), MS basic medium was most effective for inducing multiple shoot formation. Adventitious shoots (more than 40 per multiple shoot) were formed continuously on MS basal medium supplemented with 0.5 ㎎ㆍL?¹ BAP and 0.01 ㎎ㆍL?¹ NAA through four generations of subculture. Compact plantlets were induced on MS medium containing with 0.5 ㎎ㆍL?¹ paclobutrazol in vitro.
