Extraction and Purification of Peroxidase from Soybean Shell

To set up a effective purification method of soybean shell peroxidase from crop waste,and to detect its purity and molecular weight.Soybean shell peroxidase was extracted and purified by means of crushing,ultrasonic extraction,ammonium sulphate fractionated precipitation,dialysis,DEAE-Sepharose Flast Flow ion-exchange chromatography and Sephadex G-100 Gel column chromatography,using the enzyme activity and specific enzyme activity as detective markers.And its purity and molecular weight was tested through SDS-PAGE.The optimum technological conditions of extraction for SSP were:Temperature at 35℃,pH 5.2,ratio of liquid to solid at 20;when the concentration of ammonium sulphate were 50%-90%,protein content and enzyme activity were more high relatively;the molecular weight of purified SSP was 3.5×104u-4.5×104u,the value of Rz was 2.4.
