Construction and Identification of Recombinant Adeno-Associated Virus Vector Expressing VEGF_(121)

[Purpose] To construct the recombinant adeno-associated virus vector expressing VEGF121 and to provide an experimental basis for clinical research and its expression in vitro.[Methods] Human VEGF121 gene was induced by RT-PCR and cloned into pAAV-MCS vector for constructing pAAV-VEGF121 expression vector.pAAV-VEGF121 vector was identified with double digestion and DNA sequencing.[Results] RT-PCR amplified a 444bp specific electrophoretic band.The results of double digestion showed that pAAV-VEGF121 vector was successfully constructed with DNA sequencing similar to VEGF121mRNA.[Conclusion] pAAV-VEGF121 vector is successfully constructed and establishes an experimental basis for further exploring the mechanism of VEGF gene.
